The past and present lives of pig pseudorabies

Pig pseudorabies was first reported in 1813 and has since become widespread in European pig herds with the spread of intensive pig farming. Since the 1970s, China's pig farms have suffered large-scale outbreaks of pseudorabies. With the introduction of natural attenuated vaccine strain BarthaK-61 in 1979 and the development of the attenuated vaccine, the epidemic was controlled. After 1990, the epidemic situation decreased significantly, and most large-scale pig farms have basically achieved purification of pseudorabies. In this context, the state proposed to purify the disease in all breeding farms in the country by 2020.

However, at the end of 2011 and the beginning of 2012, many pig farms in North China (mainly Henan, Shandong, Hebei) that had been immunized with live vaccine of pig pseudorabies gene deletion still had suspected outbreaks of pig pseudorabies, with newborn piglets showing typical neurological symptoms and mortality approaching 100%. By comparing the gE nucleotide sequences of the newly isolated PRVS, it was found that the PRVS isolated from 2012 to now are located in a relatively independent branch (genotype II variant), and have the same molecular characteristics, and are relatively distant from the previously isolated strains.

The emergence and outbreak of Pseudorabies virus genotype II variant have brought huge economic losses to China's pig breeding industry. So is the new strain more virulent? Are there quality problems with existing vaccines? How effective is the protection against new strains?

Tian Zhijun, a researcher, proposed that the LD50 test of mice after virus isolation found that the new PRV strain (HeN1) had a high fatality rate at a low age (within 2 months of age), indicating that the virulence of the variant strain was enhanced. Bartha K61 vaccine was used to perform the attack protection experiments respectively, and it was found that the attack protection rate against HeN1 strain was only 50%, and the attack protection against S strain was still 100%, indicating that the immunogenicity of the vaccine had not changed, but Bartha k61 vaccine could only provide partial protection against newly isolated porcine pseudorabies virulence (HeN1 strain).

At the same time, Professor Wang Jichun and his team also carried out a number of protection tests, showing that Bartha K61 vaccine can produce effective protection against the mutant strain, but it needs to strengthen the immunization several times; Vaccination can effectively produce protection, but it can not completely prevent infection and detoxification, and a strong infection can form a lifelong latent infection.

It can be seen that the existing porcine pseudorabies vaccine has obvious defects in the clinical protection and detoxification of the mutant strains!



"Hua Wei Qing" glory listed

1. The source strain was a mutant strain with high production and stable passage

After the new variant strain HeN1 was inoculated into suitable cells, the virus propagated rapidly from the fourth generation, and the TCID50 of the fourth and fifth generation of the virus could reach 108/0.1mL and be stable after inoculation with the ratio of 1:1000.

2. Vaccine strain 5 gene deletion, good safety

A vaccine strain with 5 gene deletions (including TK, g, gE, Us9, Us2 and some reverse repeats) was obtained by low temperature passage and drug screening. The vaccine produced by this strain has high safety, and all sheep and pigs are alive after immunization with 108TCID50 virulence, and there is no vertical and horizontal transmission. The virus was not isolated and nucleic acid was not detected in immunized pig tissue after 10 times dose immunization.

3. Good immunogenicity, can effectively prevent PRV variant strains from detoxifying in pigs

The piglets were challenged after immunization, and the immune group was completely protected, while the control group died at least 4/5. Anal swab results showed that the virus titer in the immune group was significantly lower than that in the immune group of Bartha-K61 strain at 4 days after the challenge, and no virus was detected at 7 days after the challenge, effectively preventing the spread of PRV variant strain in pigs.

4. The latest freeze-drying process for vaccine production ensures the stability of vaccine quality

5. One immunization, protective antibodies maintained for more than 6 months
